Protein Quantitation
Protein assays use xMAP technology to measure multiple protein analytes simultaneously per well. Capture antibodies are bound to Luminex beads which are internally dyed. The conjugation of a specific antibody to a distinct bead allows for analysis of multiple analytes in a single well, e.g. IL-9 is captured via bead region #12. Samples are mixed with the bead sets. Analytes of interest within the sample are bound by the capture antibodies. Fluorescently labeled (Phycoerythrin) detection antibodies specific to the analytes of interest are added, forming an antibody-antigen sandwich. Completed assays are read on a Luminex instrument where one laser classifies the bead type to determine the analyte that is being detected while a second laser determines the magnitude of the bound analyte (PE-derived signal).
Gene expression with QuantiGene Plex assays
The Luminex instrument and bead platform is valuable for gene expression studies with QuantiGene Plex assays. This hybridization-based assay uses branched DNA (bDNA) technology for RNA transcript measurement, relying on signal amplification instead of target amplification. The assay features a simple ELISA-like workflow and works with challenging sample types such as degraded and cross-linked RNA in FFPE tissues and blood, without the need for RNA purification. It can screen up to 80 genes in a single well of a 96-well plate, maximizing data output.
